Analysis of Gallbladder Bile by Nuclear Magnetic Resonance(NMR) Spectroscopy

Abstract
Recently cholesterol gallbladder stone is increasing in Korea. In the pathogenesis of cholesterol gallstone, the supersaturation of cholesterol compared to bile acids and lecitin is important. Conventional methods of bile juice analysis such as
Gas
Chromatography or High Performance Liquid Chromatography are complex and time consuming. NMR Spectroscopy has been widelv used to analyze the molecular structure. Recently, in vovo NMR Spectroscopy is applied to the study of the alterations of
energy
metabolism.
We have obtained proton NMR Spectrum from gallbladder biles and and standard reagents including cholesterol, cholic acid, chenodeoxycholic acid, lithocholic acid, deoxycholic acid and lecithin. Also, we analyzed the chemical compositions of
gallbladder
bile by Gas Chromatography. In our study, we found out the unique peak of cholesterol at 5.3~5.4 ppm in the proton NMR Spectrum and observed good correlation between the area of unique peak and cholesterol contents obtained by Gas Chromatography.
But we
could not differentiate the peaks of each bile acid and lecithin.
In conclusion, NMR Spectroscopy will be utilized as a tool for measurement of cholesterol in gallbladder bile, but more developed NMRS Spectroscopy is required for analysis of chemical composition of gallbladder bile.
